This chapter discusses the continuous processing of pharmaceutical solid dosage forms, focusing on continuous wet granulation. Based on the advantages offered by continuous manufacturing, it highlights the drivers for the pharmaceutical industry to shift from batch to continuous processing. It reviews the techniques that allow continuous wet granulation of pharmaceuticals: fluid-bed granulation, twin-screw granulation, and ring layer granulation. For continuous twin-screw granulation – the most commonly used continuous wet granulation technique – the process parameters affecting the critical quality attributes of the granules are discussed. The currently available integrated continuous manufacturing lines with process analytical technology (PAT) technology for monitoring and control of granule quality are presented.
