Screening methods for compounds modulating polyamine transport

The invention relates to vitro methods of identifying compounds modulating (stimulating) polyamine transport into a cell comprising the step of: a) providing a first eukaryotic cell culture wherein the endogenous P5B type ATPase gene is inactivated, wherein the cells comprise an inserted copy of a wild type or a disease causing mutant of the P5B type ATPase, and providing a second eukaryotic cell culture wherein the endogenous P5B type ATPase gene is inactivated, wherein the cells comprise an inserted copy of an inactive P5B type ATPase, b) contacting the first and the second cell culture with a labelled polyamine and with a test compound, c) measuring the uptake of the labelled polyamine by the first and the second cell culture, d) determining the difference in uptake between the two cell cultures, wherein a difference in uptake between the first and the second culture, indicates that the test compound modulates polyamine transport.
