Het in kaart brengen van het anti-viroom van belangrijke pathogene RNA virussen doormiddel van multiplex beeldvorming, in vitro evolutie en chemogenomics

Mankind is constantly confronted with sudden outbreaks of deadly RNA viruses. Yet there are no drugs for the treatment or prophylaxis. In this project, we will map, using chemogenomics, the molecular targets for inhibition of selected viruses. First, multiplex assays will be developed to screen large series of molecules simultaneously against multiple viruses. We will engineer a panel of viruses that express a reporter whereby each of the reporters has a different color (a rainbow panel). The antiviral effects in mixed cultures will be quantified using our fully automated Caps-It facility. Once a panel of inhibitors is identified, we will study what their molecular target is. This will finally allow to map where this panel of inhibitors “meet their antiviral targets” and thus to develop a so-called antivirome or a map of the Achilles heel of these viruses.
