13.05 - 1,3-Diazepines

This chapter discusses the recent developments in the literature regarding 1,3-diazepines, partially hydrogenated derivatives and the corresponding fully saturated 1,3-diazepanes. Also oxo and thioxo analogues as well as benzofused and other fused derivatives are included in the discussion. The work provides a first point of entry to the progress in synthetic accessibility, characterization and reactivity of these aforementioned 1,3-diaza systems. Furthermore, attention is paid to the theoretical and experimental studies that have been performed. The interest in these structures continues to grow considerably which is reflected in the numerous X-ray structures that have been published over the past decade. With respect to this, recent developments in ring-expanded N-heterocyclic carbene chemistry are discussed in more detail. Additionally, 1,3-diazepine rings are embedded in several glycoluril-based supramolecular host systems of which main applications are highlighted. Furthermore, 1,3-diazepines continue to draw the attention of medicinal chemists and are usually considered for their structural analogy to other systems: ring-expanded nucleosides are a particular example of which many derivatives have anti-cancer and anti-viral properties. Finally, novel 1,3-diazepines based hypnotics have been developed.
