This question becomes particularly pressing when we inquire how the usual definition can be extended to cover potential, rather than actual properties. We present a statistically operational candidate for such an extension and show that its representation automatically captures some essential Hilbert space structure. If it is the nature of observation that is responsible for the Hilbert space structure, then we should be able to give examples of theories with scope outside the domain of quantum theory, that employ its basic structure, and that describe the optimal extraction of information. To address the time and space complexity issue in SampEn, we optimize Bucket-Assisted SampEn by dynamic memory allocation to avoid space complexity and accelerate the optimized Bucket-Assisted SampEn on Field Programmable Gate Arrays (FPGA). Our method accelerates Bucket-Assisted SampEn through efficient random storage and data access on FPGA. Furthermore, we introduce a scheduling strategy to handle unbalanced loads for time-intensive inter- and intra-similarity comparisons. We validate our approach on multi-source biomedical signals and demonstrate its effectiveness by achieving more than two orders of magnitude faster than straightforward defined SampEn computation. Our work provides a practical and effective approach for measuring time series complexity using Bucket-Assisted SampEn on FPGA, with the potential for real-time applications with long-term inputs." The signal's spectrum of the oscillometric waveform is in current literature badly understood or explored. The only known link between the oscillometric waveform and the blood pressure is the maximum of the oscillometry's envelope equalling the mean arterial pressure (MAP). This link is established under the assumption that the oscillometry is an AM-signal. Unfortunately, computing the MAP is difficult in practice due to the non-sinusoidal nature of the actual measured signals. Daarom is er een grote nood aan nieuwe klassen van antibacte riële middelen met een nieuw werkingsmechanisme om kruisresistentie met bestaande antibiotica te vermijden. Daarenboven vertonen bacteriën in een biofilm een verminderde antibioticagevoeligheid met als gevolg d at biofilm-gebaseerde infecties, zoals vreemdlichaam-geassocieerde infec ties, moeilijk effectief te behandelen zijn. Met dit project werd getracht een nieuwe klasse antibacteriële middelen te ontwikkelen, gericht tegen het bacteriële type I signaalpeptidase (SP ase I). Het bacteriële SPase I is een serine endopeptidase veranke rd in de cytoplasmamembraan en speelt een essentiële rol in eiwitsecreti e aangezien het verantwoordelijk is voor de katalytische afsplitsing van signaalpeptiden van secretorische eiwitten tijdens of kort na hun doorg ang doorheen de cytoplasmamembraan. De combinatie van het universe el en essentieel karakter van het doelwit, de gemakkelijke toegankelijkh eid van de actieve plaats aan de buitenkant van de cytoplasmamembraan en de mogelijkheid tot selectieve inhibitie van bacteriële enzymen, maakt van het SPase I een interessant doelwit voor de ontwikkeling van antibac teriële middelen met een nieuw werkingsmechanisme. Het doel van dit project was dan ook de functionele analyse van de SPase s I van Staphylococcus epidermidis en het verder onderzoeken van deze SP ases I als antibacterieel doelwit in biofilm-geassocieerde bacteriën in onze zoektocht naar potente SPase I inhibitoren. De opportunistisc he pathogeen S. epidermidis wordt meer en meer resistent tegen de gebrui kelijke antibacteriële behandelingswijzen en kan daarom een belangrijke bron vormen van levensbedreigende infecties geassocieerd met implantaten en biofilmen. De meeste bacteriën hebben één SPase I voor he t processen van alle secretorische eiwitten. Het genoom van S. epi dermidis bevat echter drie SPase I coderende genen, namelijk sip1, sip2 en sip3. Sip2 en sip3 coderen voor functionele SPases I terwi jl Sip1 de essentiële katalytische Lys ontbreekt en daarom verondersteld wordt inactief te zijn. Allereerst werd het SPase I onderzocht als doelwit voor biofilm-gebaseer de infecties. Genexpressiestudies toonden een initiële toename in expressie van de S. epidermidis sip genen in planktonische en in sessiel e bacteriën en duidden op een meer uitgesproken expressie in sessiele ba cteriën. Bovendien leidde de behandeling van S. epidermidis biofilmen met de gekende lipohexapeptide SPase I inhibitor arylomycine A 2 tot een antibacterieel effect vergelijkbaar met rifampicine. Ten slotte kon activiteit van het SPase I-afhankelijk gesecreteerde eiwit&nb sp;ß-lactamase aangetoond worden in het cultuurmedium van biofilm-g eassocieerde S. epidermidis cellen. Dit bevestigt de in vivo funct ionele activiteit van SPases I in S. epidermidis biofilmen. In een tweede deel werden twee in vitro testsystemen ontwikkeld om de fu nctionele activiteit van de opgezuiverde SPases I te onderzoeken en om d e activiteit van mogelijke SPase I inhibitoren te evalueren. In he t eerste testsysteem werd het processen van een natuurlijk preproteïnesu bstraat geanalyseerd, terwijl in het tweede testsysteem het splitsen van een fluorescerend synthetisch peptide gebaseerd op de sequentie van dit natuurlijke substraat werd gemeten. Met behulp van beide testsyst emen werd in vitro functionele activiteit van de opgezuiverde S. epiderm idis Sip2 en Sip3 enzymen aangetoond, terwijl Sip1 inderdaad inactief bleek te zijn. We toonden verder aan dat de laatstvermeld e FRET-gebaseerde test een snelle en gemakkelijke methode is om kwantita tieve informatie te verkrijgen over de activiteit van SPase I inhibitore n, terwijl de meer uitgebreide preproteïnetest kan dienen om de activite it van SPase I inhibitoren verder te bevestigen. Tenslotte werden peptidomimetica SPase I inhibitoren op een rationele ma nier ontwikkeld. Een substraat-gebaseerd peptide aldehyde, decanoy l-PTANA-aldehyde, inhibeerde S. epidermidis Sip2 en Sip3, S. aureus SpsB en E. coli LepB in vitro, maar vertoonde de beste activiteit tegen S. a ureus SpsB met een IC50 waarde van 0.09 µM. Deze inhibitor wa s echter maar matig antibacterieel actief in vivo. Combinatie van het peptide aldehyde met amoxicilline leidde wel tot een sterke verlagin g van de MIC van amoxicilline tegen een MRSA stam. Deze bevinding ondersteunt het idee om SPase I inhibitoren te gebruiken in combinatie m et ß-lactam antibiotica om de secretie van ß-lactama se te blokkeren en op deze manier ß-lactamase secreterende res istente stammen opnieuw gevoelig te maken. We kunnen dus concluderen dat we in dit werk aangetoond hebben dat, onda nks de lage metabolische activiteit van bacteriën in een biofilm, het ba cteriële SPase I ook van essentieel belang is voor deze bacteriën, waard oor het een veelbelovend doelwit is voor het ontwikkelen van antibacteri ële middelen tegen biofilm-gebaseerde infecties. Bovendien hebben we twee in vitro testsystemen op punt gesteld en hebben we aangetoond da t de FRET-gebaseerde test een waardevolle methode is om SPase I inhibito ren te identificeren en te valideren. "Obstructive Sleep Apnea Screening by Joint Saturation Signal Analysis and PPG-Derived Pulse Rate Oscillations" "Carolina Varon Perez, Dries Testelmans, Bertien Buyse" "Obstructive sleep apnea (OSA) is a high-prevalence disease in the general population, often underdiagnosed. The gold standard in clinical practice for its diagnosis and severity assessment is the polysomnography, although in-home approaches have been proposed in recent years to overcome its limitations. Today's ubiquitously presence of wearables may become a powerful screening tool in the general population and pulse-oximetry-based techniques could be used for early OSA diagnosis. In this work, the peripheral oxygen saturation together with the pulse-to-pulse interval (PPI) series derived from photoplethysmography (PPG) are used as inputs for OSA diagnosis. Different models are trained to classify between normal and abnormal breathing segments (binary decision), and between normal, apneic and hypopneic segments (multiclass decision). The models obtained 86.27% and 73.07% accuracy for the binary and multiclass segment classification, respectively. A novel index, the cyclic variation of the heart rate index (CVHRI), derived from PPI's spectrum, is computed on the segments containing disturbed breathing, representing the frequency of the events. CVHRI showed strong Pearson's correlation (r) with the apnea-hypopnea index (AHI) both after binary (r=0.94, p 0.001) and multiclass (r=0.91, p 0.001) segment classification. In addition, CVHRI has been used to stratify subjects with AHI higher/lower than a threshold of 5 and 15, resulting in 77.27% and 79.55% accuracy, respectively. In conclusion, patient stratification based on the combination of oxygen saturation and PPI analysis, with the addition of CVHRI, is a suitable, wearable friendly and low-cost tool for OSA screening at home." "Signal acquisition and analysis of ambulatory electromyographic recordings for the assessment of sleep bruxism: A scoping review" "Toon De Laat" "BACKGROUND: Ambulatory electromyographic (EMG) devices are increasingly being used in sleep bruxism studies. EMG signal acquisition, analysis and scoring methods vary between studies. This may impact comparability of studies and the assessment of sleep bruxism in patients. OBJECTIVES: (a) To provide an overview of EMG signal acquisition and analysis methods of recordings from limited-channel ambulatory EMG devices for the assessment of sleep bruxism; and (b) to provide an overview of outcome measures used in sleep bruxism literature utilising such devices. METHOD: A scoping review of the literature was performed. Online databases PubMed and Semantics Scholar were searched for studies published in English until 7 October 2020. Data on five categories were extracted: recording hardware, recording logistics, signal acquisition, signal analysis and sleep bruxism outcomes. RESULTS: Seventy-eight studies were included, published between 1977 and 2020. Recording hardware was generally well described. Reports of participant instructions in device handling and of dealing with failed recordings were often lacking. Basic elements of signal acquisition, for example amplifications factors, impedance and bandpass settings, and signal analysis, for example rectification, signal processing and additional filtering, were underreported. Extensive variability was found for thresholds used to characterise sleep bruxism events. Sleep bruxism outcomes varied, but typically represented frequency, duration and/or intensity of masticatory muscle activity (MMA). CONCLUSION: Adequate and standardised reporting of recording procedures is highly recommended. In future studies utilising ambulatory EMG devices, the focus may need to shift from the concept of scoring sleep bruxism events to that of scoring the whole spectrum of MMA."
